FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and ichthyopathology and can be used to produce adsorbed salmonids vibriosis vaccines. Controlled periodical process of deep cultivation of the selected production strains of halophilic vibrios is applied. Main component of the active substance of the vaccine is an inactivated suspension mixture of halophilic vibrios Listonella (Vibrio) anguillarum serotype 01 VIEV AF 3/4 and serotype 02 VEIV VBF 07, grown separately in Hottinger broth with 10 % sea water, salinity of 3.3 %, and amine nitrogen of not less than 180 % mg at 25 °C for 26 hours, pH 7.8, pO2 – 35 % of saturation with air oxygen, parallel to each other, to a maximum level of accumulation of microbial cells in medium of growth. Obtained microbial mass is treated with formalin to a final concentration of 0.37 %, then subjected to 3-fold reprecipitation with centrifugation of antigens in 0.15 M phosphate-salt buffer pH 7.6, to get rid of preserving agent and ballast components. Obtained antigens of each serotype with concentration of 1.25×109 m.cl./cm3 are taken in equal proportions, taking into account the total final concentration of 2.5×109 m.cl./cm3 in the mixture, as an adjuvant – aluminum hydroxide in 6 % of gel in volume of 12 % of total volume with further dispersion on rotary plant.
EFFECT: method enables to obtain a preparation having prolonged immunogenicity, which generally improves the quality of the end product.
