SUBSTANCE: group of inventions relates to biotechnology. Disclosed is a biosensor and a method for determining an analyte, as well as a method of making a biosensor. Biosensor comprises a substrate, a working electrode with an electroconductive area in conducting contact with the mediator layer and in the diffusion contact with the mediator layer and an enzyme layer containing the oxidase. Mediator layer is produced by electrodeposition, contains an electrocatalytic agent containing MnO2. Manufacturing method involves electrodeposition of a mediator layer on a part of a conductive platform and deposition of an enzymatic layer containing an oxidase on a part of the mediator layer. Method of determining an analyte involves bringing a sample into contact with an enzymatic layer of a biosensor, closing a working electrode, a mediator layer and a counter electrode of the electric circuit, applying voltage to the closed electrical circuit, measuring the flowing current and determining the analyte in the sample.
EFFECT: invention enables to obtain a well-connected mediator layer with underlying surface, absence of technical difficulties associated with application of said layer by screening method, obtaining low zero currents, as well as possibility of accurately setting size of working electrode and amount of deposited mediator.
