SUBSTANCE: described is a group of inventions involving a method for amplifying a sequence of a target nucleic acid with an unknown sequence (versions), method for determining a target nucleic acid sequence containing a region with an unknown sequence, and a kit for isothermal amplification of a target nucleic acid with an unknown sequence. In one embodiment, the method comprises contacting a target nucleic acid sequence with an oligonucleotide for insertion into a chain, which has non-extendable 3'-end, and at least one primer capable of amplifying the target nucleic acid sequence, wherein the oligonucleotide for chain insertion is bound to the adapter sequences to make oligonucleotide binding sites for insertion into 3'–5' and 5'–3' direction of the chain, in the target nucleic acid sequence to provide binding of at least one primer.

EFFECT: invention extends range of means for amplification of sequences of nucleic acids-targets with unknown sequence.
