FIELD: molecular biology.
SUBSTANCE: group of inventions specifically relates to oligonucleotides and a method for their use for detection and/or differentiation of target nucleic acids. A method for determination of the presence or absence of the first and the second targets in a sample includes obtaining a reaction mixture by bringing the sample or its derivative presumably containing the first and/or the second targets or their amplicons into contact with the first and the second closed oligonucleotides of a “stem-loop” type; treatment of the reaction mixture, detection of levels of the first and the second fluorescent signals at one or more temperatures, which allows for determination of the presence or absence of the specified targets in the sample. A composition for determination of the presence or absence of the first and the second targets in the sample contains the first and the second closed oligonucleotides of a “stem-loop” type.
EFFECT: group of inventions provides for expansion of possibilities of instruments for differentiation of several targets, using one type of a detector, for example, one fluorescence channel.
