Abstract RU 2730873 C2

FIELD: biotechnology.

SUBSTANCE: described is a method for determining a profile of uterus susceptibility in a female subject during an implantation window. Endometrial condition is determined in the endometrial sample by measuring, at the first stage, the state of activity of NK cells of the uterus (uNK), wherein the uNK activity state is determined by measuring the expression of IL-18 and TWEAK, determining the IL-18/Tweak ratio and comparing the obtained value with the ratio of IL-18/Tweak of a female fertile subject; where the IL-18/Tweak ratio reduced to 0.45 times relative to the value of the IL-18/Tweak ratio obtained from a female fertile subject, can indicate a hypoactive state of the endometrium, and the IL-18/Tweak ratio increased to 2 times relative to the value of the IL-18/Tweak ratio obtained from a female fertile subject may indicate a hyperactive state of the endometrium; determining, at the second stage, the state of recruiting and maturation of uNK cells, wherein the recruiting state is determined by quantitative determination of CD56+ uNK and comparing said amount with the number of CD56+ uNK cells, derived from female fertile subject, where the number of CD56+ uNK cells reduced to 10 times when counted in the field of view, can indicate a hypoactive condition of the endometrium; and the maturation state of uNK cells is determined by measuring expression of IL-15 and Fn-14, determining the ratio of IL-15/Fn-14. Obtained values are compared with ratio IL-15/Fn-14 in female fertile subject, where value of ratio IL-15/Fn14, reduced to 0.25 times relative to ratio of IL-15/Fn14, obtained from female fertile subject, can indicate hypoactive state of endometrium, and ratio of IL-15/Fn14, increased to 2.1 times relative to relation of IL-15/Fn14, obtained from female fertile subject, may indicate hyperactive state of endometrium. Thus, establishing an endometrial condition, which is characteristic for adequate or inadequate implantation readiness, where inadequate implantation readiness may be caused by absence of susceptibility corresponding to hypoactive state of endometrium or abnormal susceptibility, which corresponds to hyperactive state of endometrium. Described is a method for increasing the success rate of reproductive technology in a female subject and a method for evaluating a patient for reproductive technology, including using the described method for determining the uterus susceptibility profile. What is also presented is a kit for implementing a method for determining an uterus susceptibility profile.

EFFECT: invention enables to determine a susceptibility profile (implantation readiness) of the uterus to increase transplantation success in artificial insemination.
