FIELD: biotechnology.
SUBSTANCE: method involves extraction of DNA from the analyzed strain, PCR with specific primers and electrophoretic detection of amplification products in an agarose gel. Characterized method is based on INDEL-markers, which represent insertions-deletions of several nucleotides. Amplification is carried out with primers to each of ten INDEL-loci (RS05410, RS06765, RS11140, RS12095, RS12365, RS14755, RS16525, RS05465, RS14470 and RS16540), having two discriminated alleles. Results are recorded by electrophoretic detection; the size of the obtained fragment is determined using a molecular weight marker or allelic markers 1 and 2. Unique INDEL-genotype is established for each analyzed strain. This method can be used as an additional method of genotyping when establishing the spatial and temporal origin of the strain, determining the genesis of the outbreak, a source of infection, possible routes of bringing in brucellosis.
EFFECT: what is declared is a method for molecular-genetic typing of Brucella melitensis strains.
