FIELD: medical microbiology.
SUBSTANCE: invention relates to differentiation of Vibrio and Aeromonas bacteria by absence of arginine dihydrolase, presence of sucrose fermentation using a differentiating mixture of reagents (nutrient medium) of a certain composition, and can be used in laboratory practice when identifying a cholera vibrio. Essence of the invention lies in the fact that the culture medium is used as a differentiating mixture of reagents with preliminary preparation of the analyzed strains, then strains are sown into a differentiating mixture of reagents, which is preliminarily poured into sterile test tubes 7–8 ml and sterilized at 104 °C for 20 minutes in a steam steriliser, wherein the test tubes with the mixture are left to solidify in an inclined position so as to form a chamfered part and a "column". Cultivation of prepared cultures of analyzed strains of cholera vibrios and aeromonads is performed on a bevelled part and prick-in in a "column", then the crops are incubated in thermostat at 37 °C for 12–24 hours. Taking into account results: if skewed part of medium and "column" are colored from green to yellow, then analyzed culture is related to genus Vibrio cholerae, wherein sucrose fermentation with formation of acidic products with no fermentation of arginine; in case coloring of beveled part of differentiating mixture is from initial green color to blue, then strains are referred to Aeromonas genus due to fermentation of arginine with formation of products of an alkaline nature due to the presence of an enzyme arginine dihydrolase.
EFFECT: method provides reduced duration and volume of studies at stages of laboratory diagnostics of cholera.
