MODIFIED GENETIC CONSTRUCT FOR RECOMBINANT EXPRESSION AND CRYSTALLISATION OF HUMAN CYSLT1 RECEPTOR Russian patent published in 2020 - IPC C07K14/705 C12N15/62 C12N15/63

Abstract RU 2735281 C1

FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology, specifically to recombinant production of human leukotriene receptor type 1 (human cysteinyl leikotriene receptor 1, CysLTR1), and can be used for expression of CysLT1 receptor. Disclosed is a genetic construct with a nucleotide sequence which is optimized for expression of CysLTR1 in insect cells. Also, additional truncation of disordered C-end of CysLT1 receptor prevents formation of crystals.

EFFECT: invention provides recombinant expression in a eukaryotic system on a cell surface by combining N-terminal peptide fragments and the position of partner protein b562RIL in a third intracellular loop, which also stabilizes the protein and helps to form crystalline contacts.
