FIELD: biotechnology.

SUBSTANCE: disclosed are compositions and methods for producing adenoassociated virus (AAV) in insect cells in vitro. Recombinant baculovirus vectors contain an AAV (Cap) capsid gene expression cassette, a Rep AAV (Rep) gene expression cassette and a baculovirus homologous region (hr), located at a distance of up to nearly 4 thousand base pairs from start codon of AAV expression cassette. Levels of producing baculovirus and AAV in insect cells carrying recombinant baculovirus containing Cap, Rep and hr are higher compared to controls containing Cap and Rep, but not containing hr. In addition, the levels of producing baculovirus and AAV in insect cells infected with recombinant baculovirus containing Cap, Rep and hr are relatively stable in serial passage of cells, whereas the levels of baculovirus and AAV production decrease with serial passage of insect cells containing recombinant baculovirus containing Cap and Rep, but not containing hr.

EFFECT: disclosed are AAV production in insect cells, methods and compositions for this purpose.
