SUBSTANCE: group of inventions relates to direct immunohistochemical staining of tissue. Disclosed is a method for detecting a target analyte in a tissue, comprising: (a) contacting tissue which contains a target analyte with multiple conjugates of a polymer enzyme / primary antibody; wherein a plurality of polymer enzyme / primary antibody conjugates comprises a population of polymer enzymes having a size distribution, characterized by the number of enzyme molecules per each polymer enzyme, wherein each conjugate of the polymer enzyme / primary antibody has a molecular weight from 400 kDa to 2000 kDa, wherein step (a) is carried out for an incubation period of 5 minutes or less; (b) substantially removing conjugates of a polymer enzyme / primary antibody that do not form a complex; and (c) bringing the tissue shear into contact with the substrate of the plurality of enzyme molecules, thereby detecting the target analyte. Also disclosed is a method of treating an individual having the disease.
