METHOD FOR MANUFACTURING A CELLLESS HYDROGEL FROM WARTON'S JELLY MADE OF HUMAN UMBILICAL CORD FOR INTRA-ARTICLE APPLICATION Russian patent published in 2021 - IPC A61K35/32 A61K35/44

Abstract RU 2745995 C1

FIELD: medicine.

SUBSTANCE: invention relates to medicine, namely to a method for manufacturing a cell-free hydrogel from Wharton's jelly of the human umbilical cord. A method of manufacturing a cell-free hydrogel from Wharton's jelly of the human umbilical cord for intra-articular use in order to restore damage to the articular cartilage, including obtaining a native umbilical cord, sterilization with a hydrogen peroxide solution, its preparation, fragmentation and homogenization, decellularization with sodium hydroxide followed by its neutralization with a solution of hydrochloric acid, washing and detached cells, neutralization with sodium hydroxide solution and re-centrifugation; lyophilization, then the resulting lyophilized matrix is solubilized by exposing the resulting matrix to pepsin in a solution of hydrochloric acid, after which the medium is neutralized by titration with sodium hydroxide to physiological pH values, under certain conditions.

EFFECT: above method allows to create a full-component matrix and hydrogel from the connective tissue of the human umbilical cord using the most physiological methods, which has regenerative properties to ensure the healing of cartilage and soft tissue injuries.
