FIELD: medicine.
SUBSTANCE: invention relates to reconstructive-regenerative medicine and concerns a method for producing a collagen-containing cell-free hydrogel from human bone tissue, purified by a patented method for producing large-block bioimplants of the Lioplast trademark®. Proposed method of producing collagen-containing hydrogel from human bone tissue for repairing damaged and destroyed supporting, connective and epithelial tissues, with collagen concentration of 1.86-3.00 mg/ml, characterized by the fact that bone biomaterials made from Lioplast bioimplants are ground in a laboratory mill at room temperature, followed by extraction of the obtained mass in 30% acetic acid for 7 days at temperature of 5 °C; obtained solution is centrifuged, acid-base indicator phenol red is added and neutralized with weak alkali solution until pH=7.2-7.4, solution is cooled and bidistilled water is added to it at ratio of 1:50, mixed and centrifuged, method involves dialysis of the obtained solution using dialysis bags of 6-14 kDa, the solution is polymerized by gradually increasing temperature to 37 °C, 1% solution of ascorbic acid and 1% aqueous solution of rhamnose is added to the obtained hydrogel; the hydrogel is homogenised. Obtained hydrogel can be subjected to lyophilisation and radiation sterilization.
EFFECT: use of the invention enables to obtain a collagen-containing hydrogel from a bone allogenic material with composition close to the complex of protein components of the starting material, which is not cytotoxic.
