FIELD: chemistry.
SUBSTANCE: invention relates to the field of chemistry and pharmaceutics, namely to a method for determining the amount of adsorption of cinnarizine by liposomes, according whereto dialysis is performed in the main dialyser and the reference dialyser filled with a colloidal solution of liposomes with a mass fraction of liposomes from soy lecithin of 0.3059±0.0470%, and a solution of cinnarizine in hydrochloric acid 0.1 M or a solution of hydrochloric acid 0.1 M, respectively, at a volume ratio of the solution of cinnarizine or the solution of hydrochloric acid and the colloidal solution of liposomes of 1:1, wherein the volumes of the solutions filling the dialysers are equal. Dialysis tubes filled with equal volumes of 0.1 M hydrochloric acid solution are placed in the dialysers at a volume ratio of the dialyser and dialysis tube solutions of 10:3, a membrane with a transmission characteristic of 6 to 8 kDa is used for dialysis. Thermostating is performed at a temperature of 40°C for 24 hours, after equilibrium is reached, the solutions of the dialysis tubes of the dialysers are subjected to spectrophotometry at a wavelength of 252 nm. The value of adsorption of cinnarizine on liposomes is determined by the results of determining the equilibrium concentration in a dialysis tube with correction of optical density for lecithin passed through the dialysis membrane in the main dialysers, wherein the optical density of the dialysate from the reference dialyser is used for correction.
EFFECT: technical result consists in quantitative determination of cinnarizine on the surface of liposomes obtained from soy lecithin by a non-destructive method.
