METHOD FOR CULTIVATION OF METHANE-OXIDISING BACTERIA Russian patent published in 2021 - IPC C12N1/20 C12Q1/04 C12R1/01

Abstract RU 2755727 C2

FIELD: biotechnology.

SUBSTANCE: method for cultivation of methane-oxidising bacteria involves cultivation of microorganisms in conditions of cascade-flow cultivation in fermenters on a liquid nutrient medium enriched with air oxygen and additionally enriched with methane and sources of nitrogen, phosphorus and mineral salts, containing potassium nitrate, magnesium sulphate, calcium chloride, dibasic sodium phosphate, monobasic potassium phosphate, iron sulphate, zinc sulphate, copper sulphate, manganese chloride, cobalt chloride, nickel chloride, sodium molybdate, ethylenediaminetetraacetic acid in specified quantities, at a pH regulation of 7.0 to 6.5 simultaneously in two fermenters - seed fermenter and master fermenter. The seed fermenter therein operates in conditions of a fill and draw process brought to the exponential phase of bacterial growth, at a ratio of the drained volume to the remaining volume of the bacterial suspension within the range (1.5 to 2.5):1. The master fermenter operates until the stationary phase of bacterial growth with periodic batch introduction of bacterial suspension from the seed fermenter into the master fermenter. The cascade flow is therein controlled by a preset number of cycles of acidification of the bacterial suspension with a 3% aqueous solution of orthophosphoric acid.

EFFECT: invention provides a possibility of optimising the technology of conducting continuous processes, increase the stability of control, productivity and applicability of the method in the national economy with subsequent scaling of experimentally tested processes using established amounts of nutrient substances and mineral salts in a nutrient medium for specific bacteria.
