FIELD: biotechnology; immunology.
SUBSTANCE: present disclosure relates to the field of biotechnology and immunology and reveals a method for obtaining an expanded population of HSPC and endothelial cells (EC) suitable for HSPC transplantation. To implement the method, CD34+ HSPC is contacted with E4ORF1+ endothelial cells of the human umbilical vein (E4ORF1+ HUVEC) in a bioreactor in the initial EC/HSPC ratio. Next, HSPC is increased by cultivating HSPC and E4ORF1+ HUVEC in the bioreactor under conditions that allow HSPC to be increased during the first time period to obtain an expanded cell population containing HSPC and E4ORF1+ HUVEC in an expanded HSPC/EC ratio. Then, the expanded cell population is collected from the bioreactor. In the resulting population, HSPC and E4ORF1+ HUVEC are simultaneously present in the same environment throughout all stages of increase and collection, at no point during the method HSPC are removed from E4ORF1+ HUVEC, and they do not come into contact with additional EC. During the implementation of the method, there is no purification to remove E4ORF1+ HUVEC from HSPC. Thus, continuous interaction between HSPC and E4ORF1+ HUVEC is maintained during increase and collection stages.
EFFECT: present invention makes it possible to improve stem cell therapy and facilitate the increase and engraftment of HSPC.
