SUBSTANCE: invention relates to the field of genetic engineering and molecular biology. Proposed is a recombinant plasmid pET32v11-Flpo with the sequence SEQ ID NO: 2, ensuring synthesis of the recombinant protein 6His-S-Flpe with the sequence SEQ ID NO: 3 in Escherichia coli cells and containing the Flpo recombinase gene including cleavage sites for proteases of thrombin, enterokinase, and TEV protease of the tobacco mosaic virus. Said plasmid is therein produced by cloning cDNA of Flpo recombinase into the pET32v11 vector, and cDNA of Flpo with the N-terminal nuclear localisation sequence NLS is in the same reading frame with 6-histidine and S-epitopes and amino acid sequences of cleavage sites of thrombin, enterokinase, and TEV protease. Also proposed is a strain of Escherichia coli BL21(DE3)/pET32V11-Flpo, producing a recombinant protein 6His-S-Flpe with the sequence SEQ ID NO: 3 and produced by transforming cells of the strain Escherichia coli BL21(DE3) by the above plasmid.

EFFECT: invention provides a possibility of producing a catalytically active Flpe recombinase with a high yield.
