SUBSTANCE: invention relates to a strain of Escherichia coli producing RNA-guided endonuclease CRISPR/CPF1. The strain of Escherichia coli BL21(DE3)pLysS/pET15b-HisCpf1, producing the RNA-guided endonuclease CRISPR/CPF1 and obtained by transformation of Escherichia coli cells of strain BL21(DE3)pLysS of recombinant plasmid DNA rET15b-HisCpf1, constructed on the basis of the vector pET15b, carrying the gene of the recombinant protein Cpf1 with a length of 1281 amino acid residues, consisting of nucleases from the bacterial strain Moraxella bovis (1261 amino acids) and an auxiliary sequence of 20 amino acids long, including a histidine label and a hydrolysis site for thrombin.
EFFECT: invention provides highly efficient production of the active recombinant enzyme Cpf1.
