METHODS, COMPOSITIONS AND DEVICES FOR STORING INFORMATION Russian patent published in 2022 - IPC C12Q1/68

Abstract RU 2765308 C2

FIELD: biotechnology.

SUBSTANCE: invention relates to biotechnology, and it is a method for synthesizing charged polymer containing at least two different monomers in a nanochip, wherein the nanochip contains one or more attachment chambers containing reagents for attaching one or more monomers or oligomers to charged polymer in a buffer solution in a form with protected ends so that only one monomer or oligomer can be attached in one reaction cycle; and one or more backup chambers containing a buffer solution, but not all reagents necessary for the attachment of one or more monomers or oligomers, where chambers are separated by one or more membranes containing one or more nanopores, and where charged polymer can pass through the nanopore and cannot pass through at least one of reagents for the attachment of one or more monomers or oligomers, while the method includes a) moving the first end of charged polymer having the first end and the second end under the action of electric attraction into the attachment chamber, wherein monomers and oligomers are attached to the specified first end in a blocked form, b) moving the first end of charged polymer with added monomer or oligomer in the blocked form to the backup chamber, c) unblocking attached monomer or oligomer, and d) repeating stages a-c, where monomers or oligomers attached at the stage a) are the same or different, until the required polymer sequence is obtained, where charged polymer is DNA, monomers are nucleotides, where oligomers are oligonucleotides, and where the nanopore has a diameter of 2-20 nm.

EFFECT: obtaining compositions and devices for information storage.
