SUBSTANCE: group of inventions relates to chemistry and molecular biology. Object 1 is a kit for producing a monoclonal cluster of amplicons with a unique target locus for genotyping, comprising a flow cell comprising a substrate and first and second capture primers attached to the substrate; and a genotyping probe fluid comprising a liquid carrier and a genotyping oligonucleotide in a liquid carrier, wherein the genotyping oligonucleotide comprises a first primer sequence, a portion of an index sequence directly attached to the first primer sequence, a portion of the priming site directly attached to the portion of the index sequence, a probe sequence directly attached to the portion of the priming site, wherein the probe sequence is a target genotyping locus having a nucleic base of interest, a restriction endonuclease site and a second primer sequence that is at least partially complementary to the second capture primer. Object 2 is a method of producing a monoclonal cluster of amplicons with a unique target locus for genotyping.
EFFECT: providing genotyping of several different target loci on any surface of a flow cell, which uses clonally amplified clusters, without the need to add additional immobilisation components.
