FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, namely to the development of a serum-free environment "VNIIZZH" (Federal animal health protection center), used for the cultivation of kidney cells of the Syrian hamster VNK-21 and the production of immunogenic components of culture viruses of foot-and-mouth disease and rabies for the manufacture of vaccines. Adaptation of the transplanted suspension cell line VNK-21 to cultivation in the developed serum-free environment "VNIIZZH" is carried out with a decrease in the amount of blood serum of cattle from 5 to 0% for 6 consecutive passages. The cultivation of adapted cells of the VNK-21 line in the developed serum-free medium is carried out for 3 consecutive passages, while in 48 hours the cell concentration increases to 5.68-5.94 million/ml at a sowing concentration of 0.6-0.7 million/ml. Using the developed serum-free medium, cells of the VNK-21 line are cultured, which makes it possible to obtain FMD virus antigen with concentrations of 146S and 146S+75S in inactivated suspensions of 0.79-0.85 and 0.95-1.06, respectively, as well as rabies virus antigen with concentrations of ribonucleoprotein and glycoprotein 0.64-0.72 and 0.66-0.73, respectively, for the manufacture of safe and highly immunogenic anti-FMD and anti-rabies vaccines.
EFFECT: expansion of the range of methods for treating animals.
