FIELD: genetic engineering.
SUBSTANCE: invention relates to the field of genetic engineering, specifically to the anti-angiogenic proteins recombinant production, and can be used in medicine. Developed is the pERIG-PGS recombinant plasmid DNA, in the Escherichia coli cells ensuring the GyrA-PGS hybrid protein synthesis, containing the Mxe GyrA modified mini-intein and the Pigastin anti-angiogenic peptide, the human pigment epithelial growth factor [44-77] fragment derivative with the Pro-Gly-Pro sequence attached to the C-terminus. By the E. coli strain BL21(DE3) cells transformation with the pERIG-PGS recombinant plasmid DNA, a strain is obtained E.coli BL21(DE3)/pERIG-PGS, producing the GyrA-PGS hybrid polypeptide. Anti-angiogenic peptide Pigastin production method comprises the producer strain culturing E.coli BL21(DE3)/pERIG-PGS, destruction of the cellular biomass, the fusion protein isolation from the inclusion bodies, its autocatalytic splitting and the target peptide chromatographic purification.
EFFECT: invention allows to produce the anti-angiogenic peptide Pigastin with high yield and by simplified technology.
