FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, specifically to the construction of chimeric antigen receptors (hereinafter – CAR) and production of CAR-T-cells, which can be used in CAR-infusion therapy, using an onco-specific intracellular antigen. Antigen-binding protein is proposed, which is an antibody or its antigen-binding fragment, capable of recognizing peptide GVYDGREHTV in the complex HLA-A2-MAGE-A4 and containing polypeptide of a variable domain of a heavy chain (VH) with SEQ ID NO: 36 and polypeptide of a variable domain of a light chain (VL) with SEQ ID NO: 38. CAR is also constructed, in which the specified antigen-binding protein is located in N-end position of protein and is outside the cell, and an intracellular domain is located in C-end position of protein and is inside the cell.
EFFECT: invention provides for the production of specific scFv to peptide GVYDGREHTV from MAGE-A4, recognizing the complex MAGE-A4 peptide/HLA-A2, and their use for the production of cytotoxic CAR-T-cells, which were specifically activated for cancer target cells expressing HLA-A2/MAGE-A4.
