FIELD: biotechnology.
SUBSTANCE: group of inventions relates to chimeric antigen receptors (hereinafter – CAR) for constructed cells for adoptive therapy. CAR is proposed for use in construction of cells expressing CAR for adoptive therapy of CD19+ tumors. CAR contains (a) an antigen-binding domain containing SEQ ID NO: 27; (b) a transmembrane domain containing SEQ ID NO: 6; (c) a spacer connecting the antigen-binding domain and the transmembrane domain, and containing SEQ ID NO: 1; and (d) an intracellular signal domain containing (i) an activating cytoplasmatic signal domain containing a signal domain of a CD3-zeta chain with SEQ ID NO: 21, and (ii) a co-stimulating signal domain obtained from human CD40, containing SEQ ID NO: 12. In some aspects, a method for the construction of cells for the expression of the specified CAR, compositions containing cells, and their use in a method for the treatment of a patient with CD19+ tumor are also proposed.
EFFECT: inventions provide for increased activity, efficiency and viability of genetically constructed immune cells for use in adoptive cell therapy.
