FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology and concerns the production of preventive and diagnostic preparations used in research institutes and biological industry enterprises in the construction of biological products, in particular vaccines against animal Aujeszky’s disease. To increase the quality of the target product due to an increase in its immunogenicity during its long-term storage, a strain “Bartha ЩБК-61” of an Aujeszky’s disease virus is obtained for the manufacture of diagnostic and vaccine preparations, related to the Herpesviridae family, Alphaherpesvirinae subfamily, Varicella virus genus, deposited on 24.02.2017 in the State Virus Collection of the Institute of Virology of the Federal State Budgetary Institution “Federal Research Institute Center of Epidemiology and Microbiology named after Honorary Academician N.F. Gamalei” under No. 2882, for the manufacture of diagnostic and vaccine preparations. In a method for the production of a vaccine against animal Aujeszky’s disease, including obtaining virus-containing material from a strain of an Aujezsky’s disease virus, grown in a transferable cell culture by suspension, its inactivation and mixing with an oil adjuvant, as virus-containing material, the vaccine contains virus-containing material from a glycoprotein-deleted gE strain of the Aujeszky’s disease virus with an infectious activity of 8.0-10.0 lg TCD50/cm3, which, before mixing with the adjuvant, is inactivated by addition of 4-5% aminoethylethylenimine solution to a final concentration of 0.001-0.005% at pH of 7.6-7.8 and temperature of 35-39°C for 20-22 hours with periodic mixing, at a certain ratio of components. In addition, in the method for the production of a vaccine against animal Aujeszky’s disease, including obtaining virus-containing material from a strain of an Aujeszky’s disease virus, grown in a transferable cell culture by suspension, its inactivation and mixing with an oil adjuvant, as virus-containing material, the vaccine contains virus-containing material from a glycoprotein-deleted gE strain of an Aujeszky’s disease virus with an infectious activity of 8.0-10.0 lg TCID50/cm3, which, before mixing with the adjuvant, is inactivated by addition of 4-5% aminoethylethylenimine solution to a final concentration of 0.001-0.005% at pH of 7.6-7.8 and temperature of 35-39°C for 20-22 hours with periodic mixing, additionally contains a chitosan stabilizer at a certain ratio of components. In addition, in the method for the production of a vaccine against animal Aujeszky’s disease, an oil adjuvant ISA 201 VG or ISA 206 VG is used as an oil adjuvant. In addition, in the method for the production of a vaccine against animal Aujeszky’s disease, a transferable cell culture VNK-21/13 is used as a transferable cell culture, or a transferable cell culture VNK-21/13-13, or a primary culture of piglet kidney cells SP.
EFFECT: obtaining a vaccine against animal Aujeszky’s disease.
