FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, namely to a method for obtaining a suspension of single viable preparation cells from cellular spheroids: three-dimensional cell cultures. To implement this method, enzymatic treatment of cellular spheroids with a dissociating agent is first carried out until a suspension of single cells is obtained at a temperature of 37°C on a mini-rocker shaker with a frequency of mixing the contents of test tubes 15-20 times/min for 10 minutes. Next, the enzymatic activity of the dissociating agent is neutralized. A mixture of TrypLE Express and Accutase enzymes, taken in a 1:1 ratio, in an amount of at least 0.5 ml per at least 200 spheroids, is used as a dissociating agent.
EFFECT: present invention makes it possible to increase the yield of single viable cells while reducing the dissociation time.
