FIELD: biotechnology.
SUBSTANCE: invention relates to the field of biotechnology, namely to obtain multipotent mesenchymal stromal cells from Wharton's jelly of the umbilical cord of a newborn. The method includes preparation of a fragment of the umbilical cord of a newborn, its mechanical grinding to obtain a homogenate, enzymatic treatment of the resulting homogenate with a solution of enzymes in a ratio of 4:1 containing 0.2% of collagenase NB4, 0.005% of hyaluronidase, 10% of accutase until a viscous suspension is formed, followed by filtration of the suspension and centrifuging the filtrate to obtain a precipitate of multipotent mesenchymal stromal cells.
EFFECT: invention provides a high yield of viable cells from the umbilical cord (30-31 × 103 cells/cm2 in 5 days with a viability of up to 98%) while reducing the processing time of the material to 3-4 hours.
