FIELD: biotechnology.
SUBSTANCE: method for preparing a suspension of prostate tumour cells is developed. Method involves grinding prostate tumour tissue to a size of less than 1 mm, suspension, enzymatic dissociation of a mixture of ground tumour tissue in a DMEM medium with a solution containing collagenase, incubation, filtration, centrifugation, resuspension, characterized by that enzymatic dissociation is carried out with a solution containing collagenase in amount of 200 mcl per 2.2 ml and DNAase in amount of 50 mcg/ml of a mixture of ground tumour tissue in a DMEMmedium, incubation is carried out for 20 minutes, and filtration is carried out through a mesh filter with pore diameter of 70 mcm.
EFFECT: method provides an increase in the total yield of cells with high viability, obtaining single prostate tumour cells for subsequent sequencing, simplification and reduction of suspension preparation time due to elimination of necessity to remove erythrocytes and suspension concentration.
