FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology. A Puumala virus minigenomic system for evaluating the antiviral activity of RNA-dependent RNA polymerase inhibitors is described. Encephalomyelitis virus internal ribosome entry site (IRES). Invention makes it possible to recreate the replicative machinery of the Puumala virus in vitro. Recombinant plasmids nStem-IRES-PUUV_L, nStem-IRES-PUUV_N are intended for intracellular synthesis of viral proteins that make up the replicative machinery of the Puumala virus. The recombinant plasmid nStem-PUUV_S_DsRed is designed to evaluate the functional activity of the minigenome system.
EFFECT: invention expands the arsenal of tools for studying the Puumala virus, allowing screening of RNA-dependent RNA polymerase inhibitors, modification of viral genome segments for functional study of viral protein domains, as well as for prototyping recombinant vaccines with a controlled replication cycle.
