SUBSTANCE: invention relates to biotechnology. Described is a gene encoding an RNA-dependent RNA polymerase of the CCHF virus having a nucleotide sequence of SEQ ID NO:1 of 11838 bp in length. Also described is the recombinant plasmid pSB3delta-RdRp, designed for the integration and expression of the RNA-dependent RNA polymerase gene of the CCHF virus, has a size of 18270 bp. and contains, in accordance with the physical and genetic map, the target gene encoding the RNA-dependent RNA polymerase of the CCHF virus, having the amino acid sequence SEQ ID NO: 2 are 3945 amino acids in length, and consists of the following elements: the β-lactamase gene (coordinates from 17218 to 18078 bp) under the control of a promoter (coordinates from 18079 to 18183 bp), as a genetic marker that determines the ampicillin resistance of E. coli cells; the origin of replication ori of the plasmid vector pUC (coordinates from 16459 to 17047 bp); 5'-ITR and 3'-ITR inverted repeats with coordinates from 17 to 288 bp. and from 15990 to 16214 p.n. accordingly, they provide the integration of the plasmid region enclosed between them into the genome of mammalian cells using Sleeping Beauty (SB) transposase; promoter and enhancer CMV - provides gene expression according to claim 1 in mammalian cells (coordinates from 855 to 1438 bp); chimeric intron - chimeric intron providing efficient transcription, stability and transport of the transcript from the cell nucleus (coordinates from 1574 to 1706 bp); T7 promoter - nucleotide sequence of the T7 bacteriophage promoter required for in vitro transcription (coordinates from 1751 to 1769 bp); L protein (RdRp) - open reading frame of the gene for RNA-dependent RNA polymerase of the CCHF virus according to claim 1 (coordinates from 1783 to 13620 bp); IRES2 - internal site of encephalomyelitis virus ribosome entry, providing expression of the puromycin resistance gene (coordinates from 13854 to 14240 bp); PuroR - puromycin resistance gene, amino acids, encoding N-acetyl transferase (coordinates from 14241 to 14843 bp); bGH poly (A) signal - polyadenelation signal (coordinates from 14874 to 15098 bp).
