SUBSTANCE: invention relates to biotechnology, microbiology, and medicine, and constitutes a constructed plasmid vector containing a gene encoding an extended-spectrum β-lactamase capable of hydrolysing penicillins and cephalosporins, and intended for replication in cells of Escherichia coli and Neisseria gonorrhoeae. The plasmid contains a plasmid DNA pblaTEM-135 of N. gonorrhoeae wherein a fragment of the bla gene is cut and substituted with a similar fragment of the bla gene with the GGT→AGT mutation leading to the substitution of glycine with a series in codon 238 (Gly238Ser), allowing for the expression of extended-spectrum β-lactamase TEM-20 capable of hydrolysing penicillins and cephalosporins. Invention also discloses a method for constructing a plasmid vector.
EFFECT: possibility of obtaining a plasmid vector for assessing the resistance of N. gonorrhoeae to β-lactam antibiotics, including the products of choice for treating the gonococcal infection — III generation cephalosporins.
