FIELD: biotechnology.
SUBSTANCE: method for ion-exchange isolation of lactic acid from culture liquid is proposed, including the isolation of lactic acid in two stages using one type of anion-exchange resin - Purolite A430 in ON-form: at the first stage, acids are sorbed in a countercurrent column with a fixed sorbent layer, for this purpose a culture liquid filtrate is passed from below, while lactic and acetic acids are sorbed on Purolite A430 anionite; at the second stage, acids are eluted with a solution of sulfuric acid from the sorbent with an eluent supply from above, lactic acid comes out first, then acetic acid; eluate containing lactic acid is collected into the receiver. The total concentration of acids is controlled by acid-base titration, the concentration of lactic acid – by spectrophotometric method, the concentration of acetic acid – by the difference between the total concentration of acids and the concentration of lactic acid.
EFFECT: present invention enables to increase the degree of extraction of lactic acid from the culture fluid filtrate and the degree of separation of acids after desorption in one cycle.
