FIELD: experimental medicine.

SUBSTANCE: method for decellularization of pig dermis for reconstructive plastic surgery includes taking pig dermis with an electrodermatome; freezing of dermis samples at a temperature of -80°C followed by defrosting; processing of samples in a shaker incubator 100 revolutions °per minute in Trypsin-Versin solution at +37°C for 6 hours with the condition of replacing the solution every two hours; consecutive two-time exposure on a shaker 100 revolutions per minute with solutions of 1% Triton X-100 for 3 hours and 4% sodium deoxycholate for 3 hours; treatment in a shaker incubator 100 revolutions per minute with a solution of DNase I at + 37°C for 4 hours, and after each exposure of the solution to the sample, it is washed once in a deionized water for 10 minutes in the same mode. At the same time, dermis samples are processed in these solutions in the ratio of sample weight to solution volume - 1:10 for 22 hours, then the quality control of decontamination is carried out, the samples meet the requirements provided the DNA content is not more than 50 ng/mg of tissue.

EFFECT: present invention enables to reduce the processing time of dermis samples with chemical detergents and enzymes to 22 hours, reduces the amount of labor, saves reagents and expands the possibilities of dermis treatment.
