SUBSTANCE: proposed plasmid genetic construct pSNCA-C-3XFLAG-2XST-donor containing alpha-synuclein gene with epitopes 3×FLAG and 2×Strep-Tag II flanked by homology arms to the AAVS1 locus of the human genome. Expression of the alpha-synuclein protein is controlled by a doxycycline-driven promoter.
EFFECT: pSNCA-C-3XFLAG-2XST-donor plasmid DNA acts as a donor molecule for introducing the alpha-synuclein transgene containing epitopes 3 into the cell genome×FLAG and 2×Strep-Tag II, required for the subsequent tandem affinity purification of protein complexes interacting with alpha-synuclein, which will allow studying the interaction pattern of alpha-synuclein with various proteins and structures in human cells in synucleinopathies, in particular Parkinson's disease.
