SUBSTANCE: group of inventions relates to an adhesive composition for transdermal delivery of an active ingredient, containing the following: an active ingredient dispersed in an inert acrylate pressure-sensitive adhesive, the active ingredient being selected from tetrabenazine, deuterated tetrabenazine, or a combination thereof, while the inert acrylate sensitive to pressure adhesive is present in an amount of from 50–97% by weight, and the inert acrylate pressure-sensitive adhesive is a copolymer of 2-ethylhexyl acrylate, methyl acrylate and either one or more acrylamide monomers without functional groups selected from epoxy, -OH, -COOH and their combinations; and an inert acrylate pressure-sensitive adhesive free or substantially free of vinyl acetate, also refers to a transdermal delivery device comprising the following: a backing layer, an adhesive composition, and a release liner, also refers to a method of administering tetrabenazine, deuterated tetrabenazine, or a combination thereof to a person in need, comprising the use of an adhesive composition or transdermal delivery device on the skin of a subject, also refers to a method of inhibiting vesicular monoamine transporter isoform 2 (VMAT2) in a subject in need, also refers to a method of treating a disease or disorder mediated by vesicular monoamine transporter isoform 2 (VMAT2) in a subject in need also relates to a method of treating hyperkinetic movement disorder in a subject in need.

EFFECT: group of inventions provides an adhesive composition suitable for transdermal delivery of tetrabenazine with a therapeutically significant penetration rate of tetrabenazine with improved storage stability.
