FIELD: medicine; ophthalmology.
SUBSTANCE: invention can be used in clinical practice to replace defects in the endothelial layer of the cornea and relates to a method for obtaining a suspension of corneal endothelial cells from a human cadaveric eye. The corneal epithelium is completely removed from the surface of the human cadaveric eye with a microsurgical scraper, and a corneoscleral disc (CSD) or a corneal disc (CD) is formed from it under sterile conditions using a trephine. With the help of tweezers, the CSD or CD is held by the edge and tilted perpendicular to the sterile Petri dish, after which, using a 30 G cannula connected to a syringe filled with an intact solution for storing the cornea, a flush from the endothelial surface of the CSD or CD is carried out with the flow of the above solution over a sterile a Petri dish. Then, the resulting solution is taken from the Petri dish with the same syringe. Next, this solution is used for the second washout of endothelial cells similarly to the first one. The third flush of endothelial cells is carried out similarly to the second flush; the resulting solution is placed in a test tube. Centrifugation is carried out at 800 rpm for 5 minutes at a temperature of 37°C. The supernatant is removed with an aspirator with a sterile tip, a solution for storing the cornea is added to the sediment and the suspension is taken into a test tube, the suspension is stored at a temperature of +4°C.
EFFECT: invention provides for obtaining a suspension of endothelial cells from a human cadaveric eyeball by an accessible and easily reproducible procedure in the presence of standard laboratory equipment, which simplifies the possibility of its use.
