SUBSTANCE: invention relates to the field of medicine, namely ophthalmology. To obtain Descemet’s membrane (DM) graft with an endothelial layer, a donor preserved corneoscleral flap is placed with an endothelial layer up in the bed of a vacuum trepan. It is stained with a solution of trypan blue. Marker labels with a conditional division of the DM surface area into 8 sectors are applied to the sclera in the area of the anterior boundary ring of Schwalbe along the circumference every 45°. After one under each sector, the DM is separated from the posterior surface of the stroma in the direction of the center. With a chisel with a diameter of 8.0 or 8.5 mm, a circular notch is made on the side of the endothelium, the DM is removed outward from the circular notch. The edge of the detached DM is lifted in one of the sectors and traction to the center is performed for 180°. Then a similar manipulation is performed in the opposite sector and the DM is separated from the underlying stroma.

EFFECT: method reduces the risk of damage to the DM when it is separated from the posterior surface of the stroma, prevents the loss of a large number of endothelial cells, as well as reduces the time and volume of manipulation.
