SUBSTANCE: invention is related to technology of molecular diagnostics of myeloproliferative neoplasms. A set of reagents for the simultaneous detection of five major somatic mutations of Ph-negative myeloproliferative neoplasms in the JAK2, CALR and MPL genes and a diagnostic method based on it are proposed. A distinctive feature of the kit is the use of original combinations of specific primers and fluorescently labelled DNA probes in two reaction mixtures. The first mixture includes the following primers: JAK2-WF, JAK2-R, CALR-D, CALR-R, 515K, 515-R5 and TaqMan probes: JAK2-P, CALR-P, 515-P6. The second mixture contains the following primers: JAK2-WF, JAK2-R, CALR-R, JAK2-RM, CALR-I, 515L, 515-R9 and TaqMan probes: JAK2-P, CALR-P, 515-P10.
EFFECT: invention allows simultaneous study of five mutations in one analytical series, using only two test tubes with reaction mixtures, which leads to savings in the volume of biological sample used for analysis, reagents and analysis time.
