SUBSTANCE: invention relates to biotechnology, in particular to a plasmid construct based on vector pSK-T7 and used for in vitro synthesis of guide RNA against a fragment of sequence of 51st exon of the Dmd gene, as well as to a method for producing a line of mice containing mutation in 51st exon.
EFFECT: invention enables to create a line of genetically modified mice for simulating Duchenne muscular dystrophy and developing therapeutic approaches.
