FIELD: medicine; endocrinology and experimental biology.

SUBSTANCE: invention is intended for modeling type 2 diabetes mellitus in combination with menopause in female Wistar rats. Menopause is modeled by bilateral oophorectomy under anesthesia: the horns of the uterus with the surrounding fatty tissue, ovaries and oviducts are removed, the wound is sutured. After 2 weeks after bilateral oophorectomy, type 2 diabetes is modeled: for 4 weeks, rats are kept on unlimited consumption of high-fat food with an exchange energy of 2,690 kcal/kg, 22% of fat, 20% of protein. Next, a solution of the pancreatoprotector nicotinamide is administered intraperitoneally at a dose of 230 mg/kg, and after 15 minutes, a solution of streptozotocin pancreatotoxin at a dose of 60 mg/kg. On the 2nd and 3rd day after the administration of the drugs, the glucose level is determined at a random point by puncture of the tail vein. With glycemia ≥11.1 mmol/l, diabetes is diagnosed. At borderline glycemia values ≥6 mmol/l and <11.1 mmol/l, an oral glucose tolerance test is performed, in which the initial glucose level is determined, after which 40% glucose solution is orally administered in an amount of 3 g/kg and repeated measurements of glycemia are carried out after 15, 30 and 60 minutes after the injection of the solution. Diabetes is confirmed when the value of glycemia at any point is ≥11.1 mmol/l.

EFFECT: method provides the possibility of obtaining a model of the patient: a woman in menopause with type 2 diabetes.
