LABELED SUBUNIT VACCINE AGAINST CLASSICAL SWINE FEVER, METHOD OF ITS PREPARATION AND USE Russian patent published in 2023 - IPC A61K39/187 C12N7/00

Abstract RU 2808703 C1

FIELD: veterinary virology; biotechnology.

SUBSTANCE: group of inventions can be used in the manufacture of a product for the specific prevention of classical swine fever. A subunit labeled vaccine against classical swine fever has been proposed. The vaccine contains the following components per 1 ml of the medicinal product: antigenic material in an amount of 30 mcg containing recombinant E2 proteins of the classical swine fever virus of genotypes 1 and 2, where genotype 1 includes a fragment of the E2 gene of the Shimen strain, and genotype 2 includes a fragment of gene E2 strain Alfort-Tübingen, as pharmaceutically acceptable target additives, contains one of the following adjuvants: carbomer 971p in an amount of 2% of the volume of the medicinal product, or oil adjuvant Montanid ISA 28 in an amount of 15% of the volume of the medicinal product. The following is proposed: a strain of recombinant nuclear polyhedrosis baculovirus Autographa californica “ШАТ-22”, used to produce a vaccine; a method of producing a vaccine, including cultivating a strain of recombinant baculovirus nuclear polyhedrosis Autographa californica “ШАТ-22”, inactivating the culture liquid, determining the completeness of inactivation of the production strain, compiling a batch of the vaccine, monitoring the sterility and harmlessness of the vaccine, determining the immunogenic and antigenic activity of the vaccine. The following is proposed: a method of using the vaccine according to which the vaccine is administered intramuscularly into the area of the upper third of the neck, behind the ear according to one of the following schemes: piglets aged 40–45 days, 2.0 ml twice with an interval of 21 days; replacement gilts and sows — twice, at a dose of 2.0 ml with an interval of 21 days, so that re-vaccination occurs 14–21 days before insemination, followed by revaccination also twice before each insemination; for breeding boars 2 times a year at a dose of 2.0 ml twice with an interval of 20–30 days. The vaccine induces the formation of antibodies with a titer of virus-neutralizing antibodies to the CSF virus of at least 1:32, which reduces the risk of transmission of the pathogenic CSF virus and significantly reduces or eliminates the incidence of disease in pigs.

EFFECT: vaccine has higher efficiency and ease of production, as well as increased antigen yield.
