FIELD: analytical chemistry.
SUBSTANCE: invention relates to a method of the semi-quantitative determination of triterpene acids in an extract of coniferous woody greens. The method of semi-quantitative determination of triterpene acids in extracts of Siberian fir woody greens by liquid chromatography-mass spectrometry is characterized by the following: the extraction is carried out with acetonitrile and chromatography of the resulting solution in an anion-exchange chromatographic mode with a mixture of 0.1% solution of formic acid and acetonitrile for gradient elution in a ratio of 25: 75 at a volumetric flow rate of 1.2 ml/min, injection volume of 2 µl, column thermostat temperature of 40°C. Detection is carried out using a mass spectrometric electrospray ionization detector operating in the negative ion registration mode at a capillary voltage of 3,000 V, a nozzle voltage of 1,000 V, a scanning range of 150–550 m/z, Da. A semi-quantitative determination of the amount of triterpene acids is carried out using a calibration dependence in the range from 0.05 to 0.50 mg/ml of either ursulic or oleanoic acid.
EFFECT: achievement of group selectivity for triterpene acids, facilitating the integration of peaks to determine the sum of components, reducing analysis time, and eliminating the need to purify extracts from more hydrophobic components.
