SUBSTANCE: invention relates to a bovine transgenic animal comprising a first cell having a genome comprising a first instrument which comprises a polynucleotide encoding a Cas9 protein, and an inverted terminal repeat (ITR) sequence at 5'-end and 3'-end of said first tool, and a second cell having a genome comprising a second instrument which comprises a polynucleotide coding a guide RNA capable of specifically binding to a target site, and an ITR sequence at 5'-end and 3'-end of said second instrument, as well as to a bovine embryo. Target site is an endopolynucleotide of said animal, or an exopolynucleotide included in the genome of said animal and located between the first ITR sequence and the second ITR sequence.
EFFECT: invention provides more efficient gene manipulation using a transgenic animal or a transgenic embryo in which components of the engineered nuclease can be expressed.
