FIELD: genetic engineering.
SUBSTANCE: present invention relates to a method of producing plasmid DNA in polymer nanospheres for intranasal delivery of plasmid DNA to target tissue. 1) Obtaining histone-coated nanospheres provides electrostatic interaction with circular plasmid DNA due to the presence in the core histone molecules of specific sequences, the so-called nuclear localization sequences; 2) association of DNA with preformed nanospheres can be carried out immediately before their use, therefore, both nanospheres and circular plasmid DNA are prepared in advance and stored in freeze-dried and frozen state, respectively; 3) biodegradable dextran nanospheres with diameter of not more than 200 nm, containing circular plasmid DNA, are intended for intranasal introduction.
EFFECT: present invention provides preservation of the native structure of plasmid DNA electrostatically bound to the surface of dextrane nanospheres with a diameter of not more than 200 nm and the formation of a macromolecular complex in their composition, high reliability and simplification of the method of including plasmid DNA in the composition of nanospheres and has a number of advantages.
