FIELD: medicine; virology.
SUBSTANCE: described is a test system for detecting subtypes of influenza A and B viruses in a latex agglutination reaction or for detecting specific antibodies to these subtypes of influenza viruses in a latex agglutination inhibition reaction. Test system includes sialylated polymer microspheres created by adsorption of sialylated glycoprotein on the surface of polystyrene microspheres, which are washed with phosphate-saline buffer pH 7.4, centrifuging for 30 s at speed of 10,000 g, then to obtained precipitate of washed microspheres 1% solution of sialylated glycoprotein on phosphate-buffered saline is added to final concentration of 1.0 x 109 polystyrene microspheres per 1.0 ml of solution, then these microspheres are incubated in a solution of sialylated glycoprotein for 1 hour with constant stirring at room temperature and precipitated by centrifugation for 30 s at rate of 10,000 g, then washed with phosphate-buffered saline and resuspended in phosphate-buffered saline with final concentration of 1.0 x 108 sialylated microspheres per 1.0 ml, to stabilize the obtained suspension, a bactericidal agent is added to it and stored at a temperature of +4 °C. Invention differs from previous developments based on immobilisation of specific antibodies on microspheres by the fact that it uses a new reagent for detecting latex-agglutinating activity of any subtypes of influenza A and B viruses. Also described is a method for detecting in a latex agglutination reaction of subtypes of influenza A and B viruses using a test system, consisting in mixing 10.0 mcl of the virus suspension with 1.0 x 106 sialylated polymer microspheres and incubation of the obtained mixture for 50 minutes at a temperature of +4 °C, into which is then added an equal volume of 6% paraformaldehyde solution prepared in phosphate-buffered saline, mixture is fixed at room temperature for 10 minutes, and the fixed material is analyzed by light microscopy.
EFFECT: invention extends the range of laboratory diagnostic tools for detecting any subtypes of influenza A and B virus.
