FIELD: medicine.
SUBSTANCE: invention refers to medicine, namely to clinical microbiology. Method involves three stages: at the first stage, cultures of lactic acid bacteria recovered from periodontal pockets are cultured on MRS agar for 24 hours at temperature of 37 °C; at the second stage, the recovered isolates are cultured on a medium for lactic acid bacteria containing following per 1 l of the nutrient medium: peptone – 10 g, yeast extract – 20 g, glucose – 20 g, tween-80 – 1 g, dipotassium hydrophosphate – 2 g, sodium acetate – 5 g, triammonium citrate – 2 g, magnesium sulphate – 0.2 g, manganese sulphate tetrahydrate (MnSO4*4H2O) – 0.05 g, meat water – the rest up to 1 l, pH 6.2, for 24 hours at 37 °C; at the third stage, pure cultures are diluted to concentration of 107 CFU/ml and incubated under a film in wells of a polystyrene plate, which is placed in a CO2-incubator with a constant composition of an oxygen-free gas mixture containing 80% nitrogen, 15% hydrogen, 5% carbon dioxide, for 3–14 days at 37 °C.
EFFECT: use of the invention enables to recreate a biofilm model in a putative oral probiotic candidate.
