FIELD: medicine.
SUBSTANCE: invention relates to analysis of biological materials, particularly blood, and can be used in radiation medicine, pharmacology. A method for evaluating the efficacy of radioprotective drugs using humanised mice involves obtaining haemopoietic stem cells (HSC) from a human donor. Prior to administration of HSC immunodeficient mice are exposed to gamma-irradiation in dose of 2.5 Gy with dose rate of 0.91 Gy/min. Then HSC is injected intravenously into a lateral tail vein in amount of 30–200 thousand cells per each animal, mice are kept for at least 8 weeks. Then one of the groups—a group of protected mice—is introduced with the tested radioprotective agent. Then, all mice are exposed to gamma radiation in dose of 0.5 Gy or 1 Gy with a dose rate of 0.91 Gy/min, human cells in the bone marrow are identified and a portion of HSC of the total number of all human CD451low/CD45+ cells is measured for each animal after irradiation in a group of mice without using a radioprotective agent and in a group of protected mice. Radioprotective agent is considered to be effective if the K14/3 coefficient in the protected mice is higher than K14/3 coefficient in the unprotected mice, where the coefficient K14/3 is the ratio of the portion of HSC on 14th day after exposure to the portion of HSC of a person on the third day after exposure.
EFFECT: invention increases reliability of evaluating efficacy and safety of radioprotective drugs.
