FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and represents a method for enzymatic production of saccharide containing at least one group of N-acetylneuraminic acid, involving the following steps: a) providing at least one genetically modified Escherichia coli cell, which contains: (1) a sialic acid biosynthesis pathway containing glucosamine-6-phosphate-N-acetyltransferase, N-acetylglucosamine-6-phosphate-phosphatase and N-acetylglucosamine-2-epimerase; (2) cytidine-5'-monophospho-(CMP) N-acetylneuraminic acid synthetase; (3) heterologous sialyltransferase; (4) has deleted or disturbed expression of genes coding L-fucose isomerase, fuculokinase, N-acetylgalactosamine-6-phosphate deacetylase, glucosamine-6-phosphate deaminase, N-acetylglucosamine-6-phosphate deacetylase, N-acetylmannosamine-6-phosphate epimerase and N-acetylneuraminic acid aldolase; (5) genes lacZ and araA in said cell are inactivated; and (6) said cell comprises a gene coding phosphoenolpyruvate synthase; b) culturing said at least one genetically modified Escherichia coli cell in a fermentation broth and under conditions allowing to obtain said saccharide containing at least one group of N-acetylneuraminic acid; and optionally c) extracting said saccharide containing at least one N-acetylneuraminic acid group.
EFFECT: invention enables to efficiently obtain a saccharide containing at least one group of N-acetylneuraminic acid.
