FIELD: biotechnology.
SUBSTANCE: invention relates to a method of producing a membrane protein on a pilot or industrial scale, involving the following steps: a) expression of said membrane protein in a host organism present in an aqueous medium, b) i) release of membrane protein from host organism by mechanical lysis of cells of host organism with formation of fragments of lysed cells, including this membrane protein and ii) recovering lysed cell fragments comprising said membrane protein in form of a solid fraction by centrifugation, c) addition of detergent solution to resuspended solid fraction obtained in b) for membrane protein solubilisation, d) recovering the liquid fraction of the solubilised membrane protein obtained in c) in the form of a supernatant by centrifugation, e) chromatography of the liquid fraction for binding or retention of the membrane protein on the stationary phase and f) elution of membrane protein from stationary phase with elution buffer, to obtain said membrane protein, wherein centrifugation at step d) is carried out at 500 to 30,000 g and said method does not include ultracentrifugation of the liquid fraction obtained in d).
EFFECT: method enables to efficiently obtain relatively large amounts of membrane proteins without deterioration of the quality of the end product.
