METHOD OF PRODUCING PEPTIDE FRACTION FROM WATER-SOLUBLE PROTEINS OF MICROALGAE, HAVING ANTIBIOTIC PROPERTIES Russian patent published in 2024 - IPC C12N1/12 C12P21/00 A61K36/05 C12R1/89

Abstract RU 2824212 C1

FIELD: biotechnology.

SUBSTANCE: disclosed is a method of producing a peptide fraction from water-soluble proteins of microalgae, involving cultivation of microalgae Chlorella vulgaris for 7–8 days at 30±2°C on a sterile nutrient medium of a given composition at photosynthetically active radiation 80–140 mcmol photons/m2·s pH 6.2–8.0 suspension aeration 0.2–0.3 l/min·l sterile gas-air mixture with carbon dioxide content of 0.02–0.5%; amount of introduced inoculum of microalgae cells is 8–12% of total volume of suspension. Grown cell biomass is separated in a centrifugal force field at a separation factor of 900–1,200 for 7–15 minutes; disintegrating for 290–310 s in an ultrasonic field with power of 140–160 W per 20–30 ml of a suspension of microalgae with moisture content of 96–99%, followed by treatment with lysozyme in a given concentration for 3.5–4 hours at 37°C; extraction of intracellular water-soluble proteins is carried out with 0.15 M phosphate buffer pH 7.4 in amount of 20–30 ml per 1 g of biomass for 20–24 hours at 2–4°C; separation of the water-soluble protein fraction is carried out in the field of centrifugal forces at a separation factor of 1,500–1,700 for 10–16 minutes; treatment of the aqueous protein fraction is carried out with pepsin, taken in a given ratio at pH 2–2.2 for 3.5–4 hours; enzyme is inactivated by heating mixture to 80–90°C for 12–16 minutes, leveling pH to 7–7.2 and purifying the peptide fraction from impurities in the field of centrifugal forces at a separation factor of 1,500–1,700 for 15–25 minutes.

EFFECT: invention enables to obtain a fraction of biologically active compounds having antibiotic action on gram-positive bacteria.
