FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, in particular to a method for producing a tissue-engineered in vitro model of human head and neck squamous cell carcinoma, characterized by that a cell component of the structure, which is cultured in a basic growth medium consisting of DMEM/F12 and 10 % foetal calf serum, an immortalized line of squamous cell carcinoma of head and neck or a primary cell culture obtained from a biopsy material of tumour tissue of squamous cell carcinoma of the head and neck, or a suspension of cells isolated from a biopsy material of tumour tissue of squamous cell carcinoma of head and neck without subsequent expansion ex vivo, is combined with a matrix of the structure, in form of cryoprecipitate obtained from stored at -20 °C blood plasma by its defrosting at 3 °C and concentration of plasma proteins by 5 times after deposition at 5000g, at rate of 200 thousand living cells per 100-300 mcl of cryoprecipitate, incubated for 2 minutes at room temperature, then the total volume of the suspension with the base growth medium is adjusted to 2 ml, which are transferred into well of 24-well plate with an ultra-low adhesive coating, then the plate is placed on an orbital shaker operating at 50 rpm and located in a CO2 incubator providing standard cultivation conditions, namely 37 °C, 5% CO2 and saturated humidity, and cultivated for at least 2 days.
EFFECT: invention enables to obtain a standardized tissue-engineered in vitro model of tumour tissue capable of prolonged cultivation and can find application in biomedical research and development for searching for new approaches to treating socially significant diseases.
